Microfluidic-Assisted ZIF-Silk-Polydopamine Nanoparticles as Promising Drug Carriers for Breast Cancer Therapy

Metal–organic frameworks (MOFs) are heralded as potential nanoplatforms for biomedical applications. Zeolitic imidazolate framework-8 (ZIF-8), one of the most well known MOFs, has been widely applied a drug delivery carrier cancer therapy. However, application ZIF-8 nanoparticles therapeutic agent hindered by challenge how to control release behaviour anti-cancer zinc ions cells. In this paper, we designed microfluidic-assisted core-shell modified with silk fibroin (SF) and polydopamine (PDA) sustained curcumin (CUR) tested these in vitro various human breast We report that microfluidic rapid mixing is an efficient method precisely proportion ZIF-8, SF, PDA, CUR simply adjusting total flow rates (from 1 50 mL/min) rate ratios. Owing sufficient during nanoprecipitation, our designer CUR@ZIF-SF-PDA had desired particle size 170 nm narrow distribution (PDI: 0.08), which much smaller than produced using traditional magnetic stirrer (over 1000 nm). Moreover, properly coated SF layer successfully enhanced capability reservoir ions. Meanwhile, self-etching reaction between PDA naturally induced pH-responsive level MDA-MB-231, SK-BR-3, MCF-7 cell lines, resulting high cellular uptake efficiency, cytotoxicity, cycle arrest.
